Supplementation with calcium and short-chain fructo-oligosaccharides affects markers of bone turnover but not bone mineral density in postmenopausal women

Can supplements with prebiotic fibres positively influence bone health in type 2 diabetes? A randomised controlled crossover trial
PMID: 40471453 2025 RCT (double-blind) n = 29
Finding: A 6-week crossover of 16 g/day inulin-type fructans (Synergy1) in 29 adults with type 2 diabetes had no significant effect on calcium (p=0.324), magnesium (p=0.352), vitamin D (p=0.564), P1NP (p=0.210) or CTX-1 (p=0.438).

A combination of prebiotic short- and long-chain inulin-type fructans enhances calcium absorption and bone mineralization in young adolescents

Finding: Pubertal adolescents on 8 g/day inulin-type fructans for 1 year had greater calcium absorption (+5.9%, p=0.04 at 1 yr) and greater gains in whole-body BMD (+0.015 g/cm2, p=0.01) and bone mineral content (+35 g, p=0.03) versus placebo.

The effect of intestinal flora intervention on bone development in children: A systematic review and meta-analysis

Finding: Pooling 28 trials (n=3,615), gut-flora interventions improved BMD (SMD 0.47, 95% CI 0.28 to 0.66, p<0.001, I2=0%), but the analysis pools prebiotics together with probiotics/synbiotics so the estimate is not prebiotic-specific and evidence quality was moderate-to-very-low.
